Photolabile Opioid Derivatives of ~ - Ala ' - Leu ' - enkephalin and Their Interactions with the Opiate Receptor

چکیده
Photolabile derivatives of ~-Ala'-Leu~-enkephalin were prepared by synthetic procedures in which a 2nitro-4-azidophenyl group is linked to the terminal carboxyl group of the enkephalin by means of an ethylenediamine or ethylenediamine @-alanine spacer. These peptides bind to opiate receptors with nanomolar affinities and inhibit electrically stimulated contractions of the mouse vas deferens and adenylate cyclase activity of NG108-15 neuroblastoma x glioma hybrid cell membranes. Both inhibitions are reversed by the opiate antagonist naloxone. Photolysis of the ligands bound to rat brain membranes results in the loss of approximately 50% of the receptor sites. This decrease in receptor number is blocked by naloxone and requires light. A photolabile [3H]enkephalin derivative labels an equivalent number of sites under similar irradiation conditions.

Photolabile derivatives of D-Ala2-Leu5-enkephalin were prepared by synthetic procedures in which a 2-nitro-4-azidophenyl group is linked to the terminal carboxyl group of the enkephalin by means of an ethylenediamine or ethylenediamine beta-alanine spacer. These peptides bind to opiate receptors with nanomolar affinities and inhibit electrically stimulated contractions of the mouse vas deferens...
متن کاملDifferentiation of delta and mu opiate receptor localizations by light microscopic autoradiography.
We have observed two discrete populations of opiate receptors that are differently localized in rat brain. Morphine-like (mu) receptors, labeled by 125I-labeled [D-Ala-2MePhe4Met(O)5-ol]enkephalin, are concentrated selectively in lamina IV of the cerebral cortex, certain thalamic nuclei, and the periaqueductal grey, while delta receptors, labeled by 125I-labeled [D-Ala2-D-Leu5]enkephalin, are m...

[~-Ala~,Leu~,Cys~]Enkephalin (DALCE) is a synthetic enkephalin analog which contains a sulfhydryl group. DALCE binds with high affinity to &receptors, with moderate affinity to preceptors, and with negligible affinity to K-receptors. Pretreatment of rat brain membranes with DALCE resulted in concentrationdependent loss of &binding sites. Using 2 nM ['H][DPen2,~-Pen5]enkephalin (where Pen repres...
متن کاملDynorphin selectively augments the M-current in hippocampal CA1 neurons by an opiate receptor mechanism.
Most electrophysiological studies of opioids on hippocampal principal neurons have found indirect actions, usually through interneurons. However, our laboratory recently found reciprocal alteration of the voltage-dependent K(+) current, known as the M-current (I(M)), by kappa and delta opioid agonists in CA3 pyramidal neurons. Recent ultrastructural studies have revealed postsynaptic delta opia...
متن کاملOpiate and alpha 2-adrenoceptor responses of rat amygdaloid neurons: co-localization and interactions during withdrawal.
Interactions between neuronal responses mediated by opiate receptors and by alpha 2-adrenoceptors were characterized in the amygdala. Extracellular single-unit recordings and microiontophoresis were performed using five-barrel microelectrodes in chloral hydrate-anesthetized rats. A subpopulation of amygdaloid cells displayed inhibitory responses to morphine or D-Ala,D-Leu-enkephalin; antagonist...
